Oligodendrocyte precursor cell maturation: role of adenosine receptors

نویسندگان

چکیده

Oligodendrocyte-formed myelin sheaths allow fast synaptic transmission in the brain and their degeneration leads to demyelinating diseases such as multiple sclerosis. Remyelination requires differentiation of oligodendrocyte progenitor cells into mature oligodendrocytes but, chronic neurodegenerative disorders, remyelination fails due adverse environment. Therefore, a strategy prompt cell towards myelinating is required. The neuromodulator adenosine, its receptors (A1, A2A, A2B A3 receptors: A1R, A2AR, A2BR A3R), are crucial mediators processes. It known that A1Rs facilitate maturation migration whereas A3Rs initiates apoptosis cells. Our group research contributed field by demonstrating A2AR inhibit reducing voltage-dependent K+ currents necessary for differentiation. present review summarizes possible role adenosine receptor ligands potential therapeutic targets pathologies

Role of adenosine in oligodendrocyte precursor maturation

Differentiation and maturation of oligodendroglial cells are postnatal processes that involve specific morphological changes correlated with the expression of stage-specific surface antigens and functional voltage-gated ion channels. A small fraction of oligodendrocyte progenitor cells (OPCs) generated during development are maintained in an immature and slowly proliferative or quiescent state ...

The role of adenosine A3 receptors in cytotoxicity of the breast cancer cell lines

The nucleoside adenosine is present within cells and body fluids of all living organisms and its production, both intra- and extracellularly, is tightly coupled to energy consumption resulting in increased level of extracellular adenosine. The physiological effects of adenosine are mediated through four pharmacologically and biochemically distinct adenosine receptors (AR), i.e. A1, A2A, A2B and...

Cell Size Control and a Cell-intrinsic Maturation Program in Proliferating Oligodendrocyte Precursor Cells

We have used clonal analysis and time-lapse video recording to study the proliferative behavior of purified oligodendrocyte precursor cells isolated from the perinatal rat optic nerve growing in serum-free cultures. First, we show that the cell cycle time of precursor cells decreases with increasing concentrations of PDGF, the main mitogen for these cells, suggesting that PDGF levels may regula...

Hypoxia Alters Cell Cycle Regulatory Protein Expression and Induces Premature Maturation of Oligodendrocyte Precursor Cells

BACKGROUND Periventricular white matter injury (PWMI) is a common form of brain injury sustained by preterm infants. A major factor that predisposes to PWMI is hypoxia. Because oligodendrocytes (OLs) are responsible for myelination of axons, abnormal OL development or function may affect brain myelination. At present our understanding of the influences of hypoxia on OL development is limited. T...
